No;  you rarely need to reboot your computer to do anything at all 
(amazing how much the Windows influence makes people think there's 
something necessary or required about reboots, isn't it? :-)

What do you mean by, "Active", though?  If you expect to have a 
server listening on that port, you'll need to edit inetd.conf (or 
whatever equivalent you're using) to arrange for that server to start,
then issue a "killall -HUP inetd" command.
